The Mispillion River is a river, approximately 15 mi (24 km) long, in southern Delaware in the United States.
It rises in northern Sussex County, in several streams approximately 6 mi (10 km) southwest of Milford. It flows northeast, forming the boundary between Sussex and Kent counties. Northeast of Milford, its head of navigation, it broadens into a serpentine estuary, approximately 5 mi (8 km) long, that enters Delaware Bay approximately 15 mi (24 km) northwest of Cape Henlopen.
The Mispillion Riverwalk follows the river as it passes through Milford, extending east and west of Milford as the Mispillion River Greenway.
