Mokane is located at 38°40'34" North, 91°52'28" West (38.676041, -91.874404)Template:GR. It is located on the left bank of the Missouri River at mile 123.4. The Katy Trail, a rails-to-trails hiking/biking path, runs through the town, which stands at mile 125.
According to the United States Census Bureau, the city has a total area of 0.8 km² (0.3 mi²). 0.8 km² (0.3 mi²) of it is land and none of it is covered by water.
